It happened Friday at around 6:30 p.m. when police raided LI Foot Spa on Jericho Turnpike.
Flushing residents 44-year-old Yongli Zhang and 43-year-old Hong Li are facing multiple charges.
The Smithtown Fire Marshal also issued five violations for fire code offenses at the building.
The raid in Commack followed a sting operation on Wednesday in Nesconset at a business called Health Spa on Smithtown Boulevard.
Police say two women from Queens were arrested and charged with prostitution.
The Smithtown Fire Marshal issued the owner of the business 13 violations for fire code offenses. The Smithtown building inspector deemed the unit unsafe for business.
In both cases, police say they launched investigations after receiving numerous complaints from the community.
