The suit stems from concerns they brought up prior to the new legislative map's approval in February.
They say it favors Republican candidates and dilutes the voting power of minority communities.
Republicans have previously said the maps are fair.
A spokesperson for Nassau County Republicans declined to comment Wednesday until they could review the lawsuit.
Democrats plan to make a formal announcement Thursday in Mineola.
